Unresolved connections

The most firmly established finding concerns the final step. “Reduced translation and lower protein synthesis drive lifespan extension,” Obata says. He attributes this benefit to improved proteostasis—the cell’s ability to maintain proteins in optimal condition.

The link between Lsp2 and ribosomes is less clear. “We still need to determine why Lsp2 preferentially targets the ribosome,” Obata notes. “That part remains largely unresolved.” The team also could not demonstrate causation by restoring ribosome levels. “Ribosomal proteins are a mixture of roughly 100 distinct components,” Obata told Ars. “To manipulate ribosomal levels, you would need to upregulate or downregulate all 100 simultaneously, which is currently not technically feasible.”

Even with all details resolved, administering longevity diets to toddlers is unlikely to become feasible in the foreseeable future, mainly because mammals—including humans—lack a direct equivalent of Lsp2.

Translational challenges

Obata considers albumin and globulins the best human functional counterparts to Lsp2. Together, they constitute roughly 90 percent of blood protein. Like Lsp2, albumin is produced in large quantities, turns over rapidly, and is sensitive to dietary protein intake.

Testing whether these proteins carry nutritional memory would require a stepwise approach. “First we must perform studies in mice, where we can manipulate proteins directly, and then possibly in primates,” Obata says. “In humans, we would need to examine correlations between albumin levels and lifespan.” If albumin is confirmed as a human nutritional memory carrier, numerous details would still need clarification.

Human albumin is not particularly rich in tyrosine or phenylalanine, so any analogous mechanism would likely depend on different amino acids. “We need to identify which amino acids are key, what molecular carrier parallels Lsp2, and whether a genuine connection exists between diet and lifespan in people,” Obata says.
